Continuity mistake: In the scene where they're in the air close to the end of the movie, when Margo tries to jump to Gru's plane, Vector grabs her and points the squid launcher at Gru, then the moon hits Vector, knocking him and Margo off his ship. When Vector climbs back up to his ship, the moon's gone. It couldn't have rolled away because Vector's ship didn't move in that short time.

Continuity mistake: It's bright daylight (seemingly morning or early afternoon) when Gru goes down to the lab to reveal his plan to steal the moon. His speech is punctuated with him posing in front of the moon in the night sky, even though it was daylight just a few minutes earlier.
Continuity mistake: In the very first shot of inside the orphanage, the tape mark on the floor is at least 3 feet away from the desk. The three girls stand behind it and 2 shots later we can see they are standing about an inch away from the desk. Then when Miss Hattie stands up, the tape is now a foot away from the table.

Factual error: The Times Square Jumbotron has a ticker-tape message beneath it. Some of the letters have sections missing, presumably to represent broken bulbs. If this is the case, the missing parts would not scroll with the message, as it's only the constant switching on and off of the bulbs that gives the impression of movement.
Continuity mistake: After Gru tells the first minion "Minions assemble", that minion slides down a pole and relays the instructions to other minions. After he does this you can see a minion standing on the right of a blue tube and he jumps in, after the camera begins to pan to the left, that same minion instantly appears back at the entrance of the tube. Probably an error in the animation sequence which returned the minion to his starting position when his animation was finished.
Continuity mistake: Justin, the boy with the toy aircraft running away from his mother, towards the pyramid in the beginning of the movie, has the toy in his left hand while going up the ramp, while falling and as he begins traveling up through the air but when the angle of view changes it is in his right hand until he lands on his father.

Plot hole: When Gru and his minions step out of the vault with the shrink ray he notices Vector walking by. Even if they managed to get inside the air duct before being seen by Vector in those few short seconds they didn't have the time to close the vault door as well without being noticed.
Character mistake: After Gru tucks the girls in for the night, Dr. Nefario tells him that there are 48 hours until the rocket's scheduled launch. The rocket launch, then, should have taken place at night, around the time the girls go to bed, two days later. The rocket, however, launches during daylight hours.
Factual error: When the moon is regrowing, it destroys the ship (which is pushing it further into the sky), yet the moon is still pushed into the sky anyway, even though there isn't anything to push it. Even if only the cockpit was destroyed and the rocket's engine was still there, it still wouldn't have been strong enough to push the moon into orbit anyway.

Continuity mistake: In the scene where Gru steals the shrink ray, his minion puts it in a glass thing. The minion doesn't close the glass door on it, but when it shows Gru and his other minion laughing for victory, the glass door is closed. And the minion didn't have enough time to close the glass container thing.
Continuity mistake: In the scene where Gru goes to get Agnes, Margo, and Edith from Vector from his "lair" (after giving him the moon) he puts his face against the camera and it displays Gru as taking up the full screen. When he then punches the camera it shows only a small portion of the screen is taken up.
Continuity mistake: When Gru gets shot with all those guns at the end of his attempts to get in Vector's house before he first sees the girls, his goggles are at the top of his head. After he gets shot, the dust is not over his eyes, indicating that his goggles were over his eyes, shielding them from the dust.
Continuity mistake: When Gru asks about the carnival game, the straw on the game handler's game is pointing straight out of the cup, but when Gru agrees and pays for the game, when the game handler accepts the money and takes a sip from the cup, the straw is suddenly pointed towards his mouth.
Continuity mistake: The car was parked a few yards outside Vector's house. The girls leave after delivering the cookies and they are half way down the path. Yet a minute and 45 seconds later, they are still walking back to the car even though it was only a few yards away from Vector's house.
