In the clip "Larry and Steve", Larry mentions a store called Stewie's.
A Quahog is another name for a clam, thus all the jokes and places in town (like the drunken clam bar) the show makes reference about.
The appearances of Quagmire and Cleveland are loosely based on Bob Hope and Jesse Jackson.
The voices of Brian, Stewie and Peter (as well as other minor characters) are all voiced by the creator, Seth Macfarlane.
During the opening theme when Lois and Peter are at the piano, when the series first started the pictures on the back wall are indecipherable but as the series progresses the pictures show Stewie, Meg and Chris's faces.
In the episode where Meg gets a makeover, she has an assistant on her tour bus, Miss Swan. For all those who don't know, this is a character on Mad TV played by Alex Borstein who also voices Lois Griffen (Meg's mom).
One of the best local jokes in the show is naming the Jr. High after the very notorious Providence mayor Buddy Cianci Jr., who is currently indicted and serving time.
In the episode "Blue Harvest, part 1," Peter narrates a telling of Star Wars from his point of view. When Chris is walking through the bar, we see Bender from Futurama, Coach John McGuirk from "Home Movies," The Evil Monkey from Family Guy and The Alien from American Dad sitting in the background talking to people.
The three skyscrapers that can often be seen on the horizon behind the Griffin's house are the actual buildings One Financial Center, 50 Kennedy Plaza and the Bank of America Tower in Providence, Rhode Island.

When Peter picks up the TV guide with Luke Perry on it, if you pause look into the lower left hand corner and it says, "if you can read this Family Guy is on the air".

In the shot of the Quahog Pest Control building you see a blue grasshopper on the building. New England Pest Control in Providence, Rhode Island has the same bug on top of its building.
Fifteen Minutes of Shame (series 2)
When all the family have been replaced by actors in this episode, the actors are re-enacting the very first episode of Family Guy where Peter gets drunk and the family are eating their breakfast while Peter is lying on the table.
The King Is Dead (series 2)
When the Griffins are in the theater, in a few shots you can see the person sitting next to Meg. This is only interesting because he is killed by her in another episode.
Death Is a Bitch (series 2)
When Stewie is riding is tricycle and knocks Lois down the stairs, it is similar to the Omen when Damien uses his tricycle to knock his mom down.
During the scene with Hitler's talk show, when Hitler starts to say the show's phone number and starts shouting in German, it translates into "You're going to need a nurse".
Family Guy Viewer Mail #1 (series 3)
During the second act (Super Griffins), just as Mayor West says he's going to roll around in toxic waste, the crowd gives him a standing ovation. Watch the bottom center of the screen; a bald black man in a grey shirt grabs the butt of the woman next to him (green dress) for no reason.
Death Lives (series 3)
After Peter gets struck by lightning and Death appears, there is a shot of the sky and the clouds seem to break apart and the sun comes out. This looks remarkably like the beginning of the opening credits for "The Simpsons," another show on FOX.
You May Now Kiss The...Uh...Guy Who Receives (series 4)
Adam West's enigmatic remark, "I'm the only Mayor West that's in trouble," was an oblique reference to the late Jim West, the Religious Right former mayor of Spokane, Washington and an ardent foe of gay marriage, until his career was ended by a gay sex scandal.
North by North Quahog (series 4)
Jesus's license plate is 'WWID' which presumably stands for 'What Would I Do?' a pun on the common phrase 'What Would Jesus Do?'
Blue Harvest (series 6)
The joke about Harrison Ford being the only actor whose career wasn't destroyed by Star Wars reportedly offended Mark Hamill. After the trilogy's success, Hamill became famous as a voice actor, playing the Green Goblin on "Spider-Man" and, more prominently, the Joker on various animated "Batman" series.

Denise Crosby, who guest stars as herself, is a distant cousin of Seth MacFarlane.
