Stephenie Meyer, author of the Twilight series, got the idea for Twilight from a dream she had back in 2003. She dreamt of a girl and a boy sitting in a meadow, talking about their impossible love for each other. The girl was just a normal, human girl. The guy was outrageously beautiful, sparkly, and obviously a vampire. It is because of this single dream that A) The Twilight series came to be and B) Meyer's vampires sparkle in the sunlight.
When Bella is invited to the Cullen's house for the first time, she and Edward head for the stairway. Against the wall is a cross lying at an angle. Though they do not discuss it in the film, in the book Edward explains to Bella the history of the cross and how Carlisle acquired it. It's clear that vampires do not fear crosses.
At the end of the movie when Bella is sitting on the bench prior to prom, Jacob offers to help her since her foot is in a cast. Edward says to Bella "I leave you alone for one second and the wolves descend." This is a veiled reference to the fact that Jacob is indeed a wolf, which you find out in the second book "New Moon".
When Mike and the rest of the boys at Forks High School are trying to get Bella's attention in the lunch room, Jessica remarks that Bella is like "a shiny new toy." This is a direct line from the first chapter of Midnight Sun, a book "in the works" by Stephenie Meyer. Midnight Sun is Twilight told from Edward's perspective. The first chapter and rough draft that leaked over the Internet are posted on Stephenie Meyer's website.
In the books, Edwards is described as being Bella's "Vampire Angel". In the scenes shot in the science lab, Edward is sitting with his shoulders hunched and behind him there is a stuffed white owl. In some shots you can't see the body of the owl and it gives the impression that Edward has white wings.
For the baseball scene, on the Twilight "Special Features" DVD they discuss how the characters had to use clear plastic Christmas ornaments (the kind used for crafts) in place of actual baseballs since they didn't use gloves (vampires would not use gloves!). Then they used CGI to change its appearance and made it look like a baseball.
At the end of the movie, when the credits are rolling, you see Rob and Kristen walking and then falling on their backs in the grass in the forest. This forest scene was deleted from the movie. What happens after they fall back is that Kristen asks Rob if he really wants a taste of her blood. He says yes. She offers her finger (right hand) and puts it in his mouth. He bites it and when she reacts a little, he calls her a "fragile little human." According to the director, Catherine Hardwicke, it is Kristen's favorite scene.
During Comic Con, one of the audience members asked if anything spontaneous occurred during filming. Catherine Hardwicke responded that when Rob was doing the kissing scene, he got a little passionate and fell on the floor. You see Stephenie Meyer next to him laughing and Rob looks chagrined. Before Twilight, Emile Hirsch and Kristen Stewart worked together in a film, Into the Wild. Emile would later recommend Kristen to Twilight director Catherine Hardwicke, who had worked with Emile on Lords of Dogtown. Interestingly enough, also in that film was Nikki Reed, who would later play Rosalie in Twilight.
Stephenie Meyer (on the bonus features DVD) discusses how she wishes she had thought of putting the dream sequence in her book that is in the movie. When Bella dreams of Edward in her bedroom, he is in the room. Bella immediately sits up and switches on the light, and he is not in the room. The audience knows that Edward was in the bedroom with Bella, and Stephenie Meyer says, "Oh, I should have done that!" in the book.
